    Abstract: PURPOSE: A bidirectional promoter of sweet potato leaf curl infection DNA virus is provided to produce vector product in molecular biology field and molecular breeding industry. CONSTITUTION: A bidirectional promoter of sweet potato leaf curl infection DNA virus is denoted by the sequence of the sequence number 1. A method for identifying the promoter activity comprises: a step of isolating a site with promoter activity and analyzing cis-acting regulation site; a step of recombining promoter site by using gateway cloning to a plant expression vector for promoter analysis; a step of transforming the recombined promoter site to Agrobacteria tumefaciens strain GV3101; and a step of screening transformant.

    Abstract: PURPOSE: A plant transformant in which partitioned GFP is expressed is provided to recover the limit of silence gene development. CONSTITUTION: A plant transformant in which partitioned GFP is expressed is prepared by a recombinant vector containing GFP gene. The plant is tobacco. The recombinant vector is pGreen0229-35S:gfp vector. A method for screening an endogenous gene which promotes or suppresses gene silencing comprises: a step of isolating mRNA from GFP expression region and GFP expression silence region of the plant tranformant; and a step of comparing mRNA expression level between the GFP expression region and GFP expression silence region and selecting different gene.

    Abstract: PURPOSE: A plant transformation method using a carrier and a transformant are provided to be widely used as a core technique in biological technology. CONSTITUTION: A carrier for transformation contains at least two restriction enzyme cleavage sites which are cleaved to protrude at least one T base at 3' in order to insert a PCR product. The carrier contains at least two restriction enzyme cleavage sites which are cleaved to protrude a T base at 3' in order to insert a PCT product between a promoter and a terminator. The restriction enzyme is AhdI, Eam1105, BciVI, BmrI, HphI, Hpy188I, HpyCH4III, or MboII. The cleavage site is 5'-CTTGG-3' or 5'-CCTGG-3'. A transformant is prepared by transforming a host cell with the carrier.
