Abstract:
PURPOSE: The monoclonal antibody mAbB55 and HmAbB128 which are produced by hybridoma strains H-mAbB55 and mAbB128 showing a superior binding specificity to human apolipoprotein B and can be used for diagnosing blood circulatory organ disease. CONSTITUTION: The human apolipoprotein B-100 antibodies are injected into a mouse for immunization of the pancreas and coalesced with myeloma cell for producing hybridoma strains. The binding specificity to apolipoprotein B-100 of monoclonal antibodies, which are produced by hybridoma strains, is analyzed by Western Blatting Method and enzyme immunoassay. The potency of monoclonal antibodies and the concentration of apolipoprotein B-100, which is bound to monoclonal antibodies, are measured by enzyme immune-assay.

Prepn. of insulin-like growth factor 1 (IGF-1) (I) by transforming an E.Coli strain with an expression vector contg. cDNA encoding (I) (prepd. from mRNA extracted and isolated from human hepatocytes), the cDNA being inserted in the form of a fusion gene with another gene, followed by culture and purificn. of the resulting (I), the improvement comprises utilising an expression vector which is a plasmid having an exterokinase recognition site between the (I) gener and the other gene, and the resulting fusion protein is cleaved with enterokinase. The prod. (I) is used for controlling the blood glucose level in diabetic patients who cannot be treated with insulin, for local treatment of bone fracture and for treating congenitalor acquired growth failure patients.
Abstract:
The measuring method of apolipo-protein concentration in blood comprises the steps of separating lipid-protein complex fractions with a density of below 1.225g/ml obtained by single centrifugation of blood, or lipid-free fractions by treating the lipid protein complex fractions with organic solvent; coating the fractions on a microtiter plate or an appropriate solid surface; adding samples and anti-apolipo protein antibodies into the microtiter plate and reacting; and then measuring the apolipo-protein concentration by general ELISA method.

본 발명은 인간 혈장 아포지단백질 B-100에 결합하는 생쥐 항체의 항원 결합 절편(Fab) 부위의 중사슬과 경사슬을 암호하는 cDNA들에 관한 것이다. 이는 세포내 발현에 의해 심장 순환기 질환 등의 진단과 관련하여 혈장 아포지단백질 B-100의 농도를 측정할 수 있는 급원물질을 제공하고 또한 심장 순환기 질환 등의 치료에 관련하여 아포지단백질 B-100을 포함하고 있는 동맥경화성 지단백질을 혈중으로부터 선택적으로 제거할 수 있는 급원물질을 제공한다.
