Abstract: PURPOSE: A method for detecting bacteria causing food poisoning of fruit juice through real time PCR is provided to simply and quickly detect bacteria in fruit juice. CONSTITUTION: A method for detecting bacteria causing food poisoning of fruit juice comprises: a step of performing lysis of fruit juice through ultrasonic wave to prepare sample; a step of adding chelating resin to the sample then centrifuging; a step of purifying the sample through gel filtration column to prepare DNA template; a step of performing realtime PCR with prepared DNA template. The gel filtration column is sephadex column.

Abstract: 본 발명은 리얼 타임 PCR 기법을 이용하여 과일 주스로부터 식중독 세균을 검출하는 방법에 관한 것이다. 구체적으로, 킬레이팅 레진과 겔 여과 칼럼을 이용하여 PCR 저해물질을 신속하게 제거함과 동시에 DNA 주형을 추출하고, 추출된 DNA 주형을 이용하여 리얼 타임 PCR을 수행함으로써 과일 주스 내에 존재하는 식중독 세균을 검출하는 것을 특징으로 한다. 이에 의하면, 기존의 농화배양과정, 전기영동과정을 거치지 않고서도 간편하고 신속하게 과일 주스 내의 식중독 세균을 검출할 수 있는 장점이 있다.

이때, 중합효소연쇄반응에 사용하는 PCR 프라이머의 염기서열은 서열목록 1과 2인 것을 특징으로 한다. 또한 이때, 탐침자(capture probe)의 염기서열은 서열목록 3인 것을 특징으로 한다.
본 발명에 따른 검출검사에 소요되는 시간은 총 5시간 미만이며 휴대가 가능하고 검사절차가 간편하며 검사결과를 육안으로 관찰할 수 있기 때문에 현장 검사용으로도 유용하다 하겠다.
