Abstract: The d- and l-isomers of 4-oxa-5-(N-methylcarbamoyloxy)-tricyclo 5,2,1,02,6 dec-8-en-3-one (I) are new cpds. Cpds. (I) have analgesic activity; e.g. d-(I) has an ED50 of 14.4 mg/kg (p.o., mouse) in the writhing test, compared with 21.6 for the racemate and 232 for l-(I). The optical isomers can be prepd. by reacting dl-4-oxa-5-hydroxy-tricycloc 5,2,1,02,6 dec-8-en-3-one (II) with d- or 1-3-aminomethylpinane (III), separating the resulting diastereomers (IV) by fractional crystn., cleaving the purified diastereomers with acid to give optically active (II), and reacting this with MeNCO or MeNHCOCl to give optically active (I).
