PROBLEM TO BE SOLVED: To provide a sufficient amount of a recombinant active heterodimer- AMV-RT. SOLUTION: An active heterodimer-AMV-RT is produced in a prokaryotic host cell by (i) cloning one or more DNA sequences encoding the α-chain and/or the β-chain of AMV-RT to an expression plasmid, (ii) transforming the expressed plasmid to a prokaryotic cell, (iii) inducing a soluble expression of heterodimer-AMV-RT and (iv) isolating the recombinant heterodimer-AMV- RT from the cell.

SOLUTION: The transferase is a truncated TdT derivative wherein at most 161 amino acids at the N-terminal are truncated and exhibiting enzymatic activity in a solution containing Co 2+ ion corresponding to 20-30 times the enzymatic activity of natural terminal deoxynucleotidyl transferase (TdT). Una solución acuosa en la que está ausente un agente reductor con un grupo tiol, comprendiendo la solución acuosa un polipéptido, comprendiendo el polipéptido una variante de la T7 ARN polimerasa (variante de T7), teniendo la variante de T7 las propiedades de (i) actividad de ARN polimerasa dependiente de ADN, y (ii) una composición distinta de aminoácidos en comparación con el polipéptido de 883 aminoácidos de la T7 ARN polimerasa de SEC ID Nº: 2 (referencia de tipo silvestre), en la que en una posición seleccionada de la 510 a la 530 de la variante de T7, numerada a partir del extremo N-terminal de la referencia de tipo silvestre, está presente un resto de Cisteína, en la que la variante de T7 comprende una o más sustituciones de aminoácidos de las cuales una (o la primera) está en la posición 723 y donde la Serina sustituye al resto de Cisteína (Cys723Ser), y en la que está ausente la formación de homomultímeros en la solución acuosa de la variante de T7 como resultado de uno o más enlaces disulfuro intermoleculares.

The present invention provides improved variants of T7 RNA polymerase by introducing novel mutations which lead to improved thermostability of the enzyme. According to the invention, amino acid substitutions at the positions Val426, Ser633, Val650, Thr654, Ala702, Val795, and combinations thereof are advantageous.

The present invention further provides a method of stabilizing an aqueous composition comprising a protein with enzymatic activity of alpha-galactosidase, and a method of preparing a purified aqueous composition comprising said protein with enzymatic activity of alpha-galactosidase.

The heterologous expression of the reverse transcriptase from the Avian Myeloblastosis Virus (AMV-RT) in prokaryotic cells and in particular Escherichia coli (E. coli) is described in the present invention. The invention also includes certain measures to simplify the purification of the heterodimeric AMV-RT.
