Abstract:
PROBLEM TO BE SOLVED: To provide an improved medium for culturing Clostridium histolyticum and culture supernatants for biotechnologically producing collagenase enzymes. SOLUTION: The culture media contain water, fish gelatin and peptones derived from a non-mammalian source. The media preferably contain peptones derived from plants as a non-mammalian source. The media also may contain two or more different peptones. Disclosed are sterilized compositions comprising the media, a culture supernatant containing the collagenase of Clostridium histolyticum, a method for producing the culture supernatant, and a method for producing the collagenase. SOLUTION: The transferase is a truncated TdT derivative wherein at most 161 amino acids at the N-terminal are truncated and exhibiting enzymatic activity in a solution containing Co 2+ ion corresponding to 20-30 times the enzymatic activity of natural terminal deoxynucleotidyl transferase (TdT). Un medio líquido para el cultivo de Clostridium histolyticum, comprendiendo el medio agua, una gelatina de pescado y una peptona vegetal, seleccionándose la fuente de la peptona del grupo que consiste en soja, haba, guisante, patata, y una mezcla de los mismos, en el cual la concentración de agregado de la peptona o peptonas vegetales en el medio es entre aproximadamente el 2 % y aproximadamente el 10 % en peso por volumen, y por lo cual en el medio la concentración de gelatina de pescado es entre aproximadamente el 2 % y aproximadamente el 10 %, indicando el porcentaje peso por volumen cuando al gelatina de pescado aislada es materia seca y volumen por volumen cuando la gelatina de pescado aislada es materia líquida.
