Abstract:
A method of preparing an antigen-immobilized immuno-fluorescence slide, the method comprising: immobilizing a C-reactive protein on a slide to prepare a protein chip; mixing an antibody that specifically binds to a target protein, with streptavidin to label the antibody with a fluorescent nanoparticle; immuno-reacting the antibody by competitive mixing, assaying with a fluorescence camera, wherein the immobilizing of the C-reactive protein on the slide comprises: modifying the slide with 3-aminopropyltrimethoxysilane to prepare a modified slide; hydrating the slide modified with 3-aminopropyltrimethoxysilane; activating the modified slide by using a glutaraldehyde solution; dissolving a C-reactive protein at a concentration of 0.01-0.5 mg/ml in a 30-70 mM phosphate buffer solution (pH 6.5-7.8) to prepare an antigen solution for immobilization; placing a petri dish comprising the slide on a spotting guide and spotting 1-100 µ„“ of the antigen solution on spotting points; and performing a reaction on the slide prepared as described above for 1-6 hours to immobilize the antigen, and an immune-fluorescence slide prepared by using the method.
Abstract:
PURPOSE: A method for preparing metallothionein(MT) which is a biomarker for heavy metal pollution is provided to detect pollution of fishes. CONSTITUTION: A method for preparing metallothionein(MT) comprises: a step of injecting a heavy metal salt solution to fishes; a step of treating heating wound at the infection site and growing the fishes to induce high concentration of metallothionein into the blood; a step of collecting blood from the fishes and adding protease inhibitor to obtain serum; and a step of performing anion exchange chromatography. The heavy metal is cadmium chloride or copper sulfate.

본 발명은 표면 플라즈몬 공명을 이용하여 튀김유의 극성 분자 함량을 신속 측정하는 방법에 관한 것으로서, 측정 시료를 에탄올과 1:1로 혼합하고, 혼합 용액 중에서 에탄올 용해성 물질이 포함된 상등액을 분취하고, 분취된 용액을 대상으로 표면 플라즈몬 공명(SPR)을 이용하여 SPR 신호값을 획득하고, SPR 신호값을 극성분자의 함량으로 환산하는 환산식에 의해 튀김유에 함유되어 있는 극성 분자의 함량을 신속하게 측정함으로써 준실시간으로 튀김유의 극성분자함량을 정량 측정하는 장점을 갖는 것이다.
Abstract:
PURPOSE: A piezoelectric quartz crystal sensor for detecting food poisoning bacteria and its preparation method are provided, to improve the sensitivity and the stability. CONSTITUTION: The method comprises the steps of dissolving antibacterial antibody against food poisoning bacteria in standard buffering solution in 0.1-7 mg/ml; making 10-70 mM of thiolated cross-linking solution; putting the two solution test tube and reacting the mixture at 10-50°C for 10-300 min; adding dithiothreitol to the test tube and reacting it for 10-200 min; coating the one surface of gold electrode of pre-activated piezoelectric quartz crystal with the obtained thiolated antibacterial antibody solution; reacting it at 15-40°C for 10-300 min; drying it; and washing it with standard buffering solution and distilled water to prepare the piezoelectric quartz crystal sensor for detecting food poisoning bacteria.
