    Abstract: PURPOSE:Gamma-linoleic acid or its ester is added to conventional powder or liquid milk to give a high-quality product having nutrient composition similar to mother's milk whereby health troubles which have been observed, when conventional milk is given, are prevented. CONSTITUTION:Gamma-linoleic acid or its ester is added to powdered milk in an amount of 0.01-1wt% on the solid basis or to liquid milk in an amount of 0.001-0.1wt%. The alkyl group of the ester is preferably methyl or ethyl. In order to admix the gamma-linoleic acid or its ester to powdered or liquid milk uniformly in the stabilized state, the acid or ester is preferably clathrated and the clathrate is added thereto.

    Abstract: PURPOSE:To obtain a hepatic function improving agent exhibiting hepatocyte activating and proliferating activity by using a human-originated cornified epidermal cell proliferation factor as an active component. CONSTITUTION:The improving agent contains 0.1-0.5wt.% of a cornified epidermal cell proliferation factor as an active component. The factor is a proteinous proliferation factor refined by using a proliferation activity from a human fetus fibroblast cell culture liquid to a cornified epidermal cell of mouse as an index. The agent is properly compounded with ordinary carrier for pharmaceutical preparation and formed in the form of injection, peroral drug or enteral administration drug. It is administered at a daily dose of 1-100mg for adult. The agent exhibits hepatocyte proliferation stimulating activity higher than known hepatocyte proliferation factor without causing the risk of the aggravation of a highly fibrillated state such as hepatic cirrhosis.

    Abstract: PURPOSE:To make it possible to collect efficiently gamma-linolenic acid, by separating phosphatidylcholine having high content of gamma-linolenic acid from lipid collected from a specific mold and concentrating the phosphatidylcholine. CONSTITUTION:A strain such as Mortierella isabellina, Mortierella vinacea Mortierella ramaniana, Mortierella ramaniana var. anglispora, or Mortierella nana, belonging to the genus Mortierella is cultivated in a medium containing a highly concentrated carbohydrate as a carbon source, a mold with high lipid content is obtained and gamma-linolenic acid is produced from lipid collected from the mold. In producing gamma-linoleic acid, phosphatidylcholine having high content of gamma-linolenic acid is separated from the lipid, concentrated and gamma-linolenic acid is obtained. To separate the phosphatidylcholine, the lipid collected from the mold is subjected to alcohol extraction at the first step of multi-stage extraction.

    Abstract: PROBLEM TO BE SOLVED: To obtain a cDNA library of mutants having introduced amino acid(s) at an arbitrary site in a protein by incorporating cDNA coding for a protein into a cyclic plasmid, cleaving the obtained plasmid at an arbitrary site, followed by introducing DNA. SOLUTION: A library of cDNA is obtained which codes for a protein mutant having an introduced amino acid or peptide at an arbitrary site in its primary structure of a protein and allows selection of a desired protein mutant by incorporating cDNA coding for a protein into a cyclic plasmid, cleaving the obtained cyclic plasmid at an arbitrary site in its primary structure, inserting DNA coding for an amino acid or peptide into the cleaved site of the plasmid, wherein a nucleotide is added to 5'- and/or 3'-end(s) of DNA coding for the amino acid or peptide so as to be able to obtain a clone in which the frame of the amino acid or peptide matches with the frame of the protein into which it is introduced, and allows selecting a desired protein mutant.

    Abstract: NEW MATERIAL:A hepatic cell growth factor consisting of a substance having the following physiochemical properties and physiological activity. The substance has a mol.wt. of approximately 18,000 by a SDS-polyacrylic amide gel electrophoresis, is adsorbed on heparin sepharose and eluted in a NaCl concentration of 1.9-2,0 M and has a hepatic cell growth activity. USE:A remedy for hepatic diseases such as acute hepatitis, fulminant hepatitis, chronic hepatitis and cirrhosis. PREPARATION:For example, a spleen is cut off from a cattle, ground in a buffer solution and centrifuged. The supernatant is dialyzed in a 10mM phosphoric acid buffer solution and subjected to an ion exchange column chromatography. The non-adsorbed fractions are treated with an immobilized heparin column and adsorbed substances are eluted. An active fraction is applied to a hydroxy apatite column, and adsorbed substances are eluted. The eluted substances are treated with a heparin-immobilized column and the adsorbed substance is eluted to provide the hepatic cell growth factor.

    Abstract: PURPOSE:To exclusively produce the titled substance in high conversion, by treating gamma-linolenic acid with soybean lipoxygenase in a system of an organic solvent and an aqueous solution in the presence of an anionic surfactant. CONSTITUTION:gamma-Linolenic acid is dissolved in a binary phase system composed of an organic solvent (e.g., hexane, heptane, etc.) and an aqueous solution. The aqueous solution is a boric acid buffer solution of 6.5pH. gamma-Linolenic acid is treated with soybean lipoxygenase in the above binary phase system in the presence of an anionic surfactant such as sodium polyoxyethylene alkyl ether acetate, sarcosinate L-N, etc., to convert the gamma-linolenic acid to 9-hydroperoxy-gamma- linolenic acid. The objective product is recovered by adding hydrochloric acid to the reaction mixture, extracting the oil-soluble fraction with ethyl acetate, subjecting the extract to dehydrochlorination and dehydration and developing by a reversed phase thin-layer chromatography.
