Abstract: Preparing carotenoids (I) or their precursors (Ia) using genetically modified organisms of the genus Blakeslea, is new. Preparing carotenoids (I) or their precursors (Ia) using genetically modified organisms of the genus Blakesleacomprises: (1) transforming at least one cell; (2) optionally homokaryotizing the cells so that cells are produced in which the nuclei are all simultaneously altered in one or more genetic characteristics and these alterations are expressed; (3) selection, multiplication and culture of the modified cell(s); and (4) recovery of (I) and (Ia) from the cells. Independent claims are also included for: (1) producing at least one high-purity (I) and a nutrient, consisting of (I)-producing organisms and at least one (I), by culturing a (I)-producing Blakeslea; and (2) human and animal foods or food supplements produced by the new method.

Abstract: Cis-6-hexadecenoic acid is prepared by reacting a triphenylphosphonium salt with decanal or other aldehyde in a Wittig reaction, and saponifying the resulting ester. Preparation of cis-6-hexadecenoic acid of formula COOH-(CH2)4-CH=CH-(CH2)8-CH3 involves: (a) reacting either: (i) a triphenylphosphonium salt of formula R OOC-(CH2)5-P(R )3 X with decanal of formula CH3-(CH2)8-CH=O, or (ii) a triphenylphosphonium salt of formula CH3-(CH2)9-P(R )3 X with an aldehyde of formula R OOC-(CH2)4-CH=O in a Wittig reaction; and (b) saponifying the resulting ester of formula R OOC-(CH2)4-CH=CH-(CH2)8-CH3. R = 1-12C alkyl or aryl; R = aryl; X = anion equivalent of (in)organic acid.
