Abstract: NEW MATERIAL:Recombinant plasmid pTPGIF2 having a size of 4,660 base pairs and a structure obtained by inserting a DNA having a size of 52 base pairs and containing a sequence coding somatostatin into a BamHI incision site of a plasmid vector pTP70-1 capable of fusing a heteropeptide to a carboxy- terminal of a dihydrofolic acid reductase gene of E.coli. USE:Production of fused protein containing somatostatin. PREPARATION:A somatostatin gene is integrated into a plasmid vector pTP70-1. pTPGIF2 is kept in stable state when introduced into E.coli. C600 strain. The E.coli. C600 strain containing pTPGIF2 is deposited in Fermentation Research Institute as FERM BP-1577.

Abstract: PURPOSE:To reduce the load of a water supply system and to decrease laminar film resistance by discharging exhaust gas in the vicinity of an interface between a gas separation membrane and water, and utilizing the movement of the bubbles generated at this time. CONSTITUTION:A hollow yarn-based gas separation membrane 3 is fixed in the housing 1 of the device for recovering oxygen dissolved in water by means of the upper and lower fixing layer 2 consisting of an adhesive. The exhaust gas used in combustion, respiration, etc., is discharged through a diffusion plate 5, and sent up as bubbles 10 in a passage 8 between membranes while accompanying and circulating the water from an outlet 4. The laminar film resistance on the surface of the gas separation membrane 3 is simultaneously removed, and the exhaust gas is discharged from an outlet 9 along with the water 12. Meantime, the oxygen and other gases dissolved in water is passed through the gas separation membrane 3, recovered, and discharged from a line 7. By this method, the device can be miniaturized, and energy can be saved.

Abstract: PURPOSE:To provide an impregnant which can seal pores of an inorganic material, such as metallic casting or sinter, in a relatively short time at a low temp. and which comprises, as main ingredients, a particular polymerizable vinyl monomer and a cyclodextrin inclusion compd. of a polymerization initiator. CONSTITUTION:An impregnant for a porous material mainly composed of a polymerizable vinyl monomer contg. (meth)acrylic ester and a cyclodextrin inclusion compd. prepd. by treating a polymerization initiator (e.g., a peroxide or azonitrile initiator) with cyclodextrin. This impregnant prolongs its storage file and working file through inclusion of a polymerization initiator, and heating of the inclusion compd. causes the polymerization initiator to be released therefrom, the inclusion compd. to be decomposed, and polymerization to be initiated, so that the impregnant can be cured in a relatively short time at a low temp. The pores of a material to be treated can be sealed by filling the pores with the impregnant, washing the surface of the material with water to remove excessive impregnant, and treating the material with hot water or hot air to cure the impregnant present in the pores.

Abstract: PURPOSE:To produce a high-hardness plastic molding having a two-dimensional covalent bond-containing network structure, being highly crystalline and excellent in hardness and modulus, by polymerizing a diacetylenic nylon salt compound by heating in a mold under a high pressure. CONSTITUTION:A high-hardness plastic molding is produced by packing a diacetylenic nylon salt compound of formula I [wherein R1 and R2 are each a (substituted)alkylene, cycloalkylene or arylene] into a mold of any desired shape and polymerizing it by heating at 100-400 deg.C and 0.5X10 -15X10 atm. A highly crystalline plastic molding in the form of a two-dimensional covalent bond-containing network can be easily produced. This molding has excellent strength and modulus and such a high hardness as to be equivalent to that of a metal, so that it is very useful as a light high-hardness material. The compound of formula I can be quantitatively obtained as a crystalline form by mixing equimolar amounts of a diacetylenecarboxylic acid of formula II (wherein R1 is as defined above) and a diamine of formula III (wherein R2 is as defined above).

Abstract: PURPOSE:To promote the reaction of a polymer and a compound using minimum amount of water necessary for the processing treatment by impregnating a fiber with a compound reactive with the fiber constituting polymer, laminating with a hydrated polymer gel sheet capable of reversibly sucking and releasing water by temperature change and heating the laminate at a temperature above the phase-transition temperature of said gel. CONSTITUTION:For example, in the thinning processing of a polyester fiber with sodium hydroxide or cotton fiber with cellulase, the fiber is impregnated with the compound reactive with the fiber constituting polymer and subjected e.g. to intermediate drying. The treated fiber is laminated with a hydrated polymer gel sheet capable of reversibly sucking and releasing water by temperature change (e.g. crosslinked gel of polyvinyl methyl ether) and the laminate is sealed, heated at a temperature above the phasetransition temperature of the polymer gel and maintained at the temperature. Water is released from the gel by this treatment to promote the reaction of the polymer and the compound in moistened state. The use of the gel is effective in preventing the waste of large amount of water in contrast to conventional process and enables the supply of necessary minimum amount of water in high efficiency to perform an energy-saving processing of the fiber.

PREPARATION:For example, a DNA fragment prepared by cleaving a plasmid pMEK2 for producing leucine enkephalin with a restriction enzyme is linked to a DNA fragment obtained by cleaving a recombinant plasmid pGRF2-15 capable of coding DHFR and an amino acid sequence of the 1st to 29th residues of a bovine GRF using a T4-DNA ligase, inserted into Escherichia coli and cultured to select the resultant colony. Thereby, the objective pGRF44-22 is obtained.

Abstract: PURPOSE:To obtain a recombinant plasmid pENDC1 being a recombinant plasmid having 4676 pair of bases and capable of producing gamma-endorphin of morphine like peptide having high analgesic activity when Escherichia coli is transformed by the above-mentioned plasmid. CONSTITUTION:Gene coding gamma-endorphin expressed by the formula is synthesized and the synthesized gene is integrated into a plasmid pMEK2 to prepare a fused gene (pENDC1) of gamma-endorphin gene and dihydrofolic acid reducing enzyme (DHFR) gene, by which Escherichia coli is then transformed. DHFR-gamma-endorphin fused protein is produced by culturing the transformed Escherichia coli and then the fused protein is digested by a proteolytic enzyme to separate gamma- endorphin and the gamma-endorphin is purified.
