Abstract: Prepn. of 2,5-diketo-gluconic acid (2,5-DGA) comprises fermentive oxidn. in the presence of oxygen by (a) synthesising 2,5-DGA from glucose in a main reactor and by (b) further oxidn. of (a) to form more 2,5-DGA in a subsequent reactor without the addn. of fresh glucose until no substantial quantities of 2-keto-D-gulonic acid and/or gluconic acid are present in the end prod.. Pref. the continuous fermentive oxidn. is with Erwinia in a main reactor contg. 100 to 400g glucose/l. The pref. exchange rate is 0.05 to 0.5 litre and hour more pref. 0.1 to 0.2 litre an hour. USE/ADVANTAGE - 2,5-DGA is a precursor of 2-keto-L-gulonic acid which can be rearranged to form ascorbic acid. Currently, only batch processes have been described for the formation of 2,5-DGA. The new method is a continuous process that is faster and needs less space than known methods, and a pure prod. can be produced. In contrast to prior art methods, with a reactor in series (b), the side prod. formation of gluconic acids and 2-keto-D-gluconic acid can be avoided.
