    Abstract: PROBLEM TO BE SOLVED: To obtain a new nucleotide having a specific base sequence or such a sequence that part of the base(s) on the 5' side, 3' side or the sides other than these ends are deleted from or added to the above base sequence, and useful as e.g. a primer for identifying Chlamydia microbial species by PCR process. SOLUTION: This new nucleotide has a base sequence of the formula, or is obtained by deleting one or two bases from the end of the 5' side or 3' side of the sequence, adding -T or -TA to the end above, substituting other base(s) for one or two bases on sides other than the above-mentioned both sides or modifying at least one of these bases. This new nucleotide is useful as e.g. a primer for identifying Chlamydia microbial species by making use of PCR process. This new nucleotide is obtained by chemical synthesis of part of the outer membrane protein gene of a kind of Chlamydia microbe (e.g. Chlamydia trachomatis), and the objective identification is carried out based on the restriction enzyme digestive pattern of the amplified product by PCR process using this nucleotide.

    Abstract: PURPOSE:To differentiate the serum type by conducting a PCR reaction using a part of a gene coding the main outer membrane protein of Chlamydia trachomatis as a primer, searching the amplified product from the reaction solution, digesting it and comparing the resultant substances. CONSTITUTION:A PCR reaction is conducted by using a nucleotide represented by formulae I to IV, etc., or a sequence obtained by applying one or more modifications thereto, e.g. eliminating one or two bases from the 5' side terminal, eliminating one or two bases from the 3' side terminal, adding G or GG to the terminal or substituting one or two bases except both the terminal with another base among A, C, G, T, etc., and composed of a part of a gene coding the main adventitial protein of Chlamydia trachomatis as a primer. The amplified product derived from this gene is searched out from the reaction solution and the searched out product is digested by a restriction enzyme. The size of the DNA fragment of the digested material is analyzed and compared, thus differentiating the serum type of a microorganism, Chlamydia trachomatis.
