Abstract:
PURPOSE: A pharmaceutical composition containing glyoxalase fusion protein is provided to prevent and treat neurodegenerative disease such as brain ischemia. CONSTITUTION: A glyoxalase fusion protein comprises 9-15 amino acid residues. The cell permeability efficiency is improved by covalent bond of a transport domain to the end of glyoxalase. The transport domain is HIV Tat 49-57 residu, oligolysine, oligoarginine, or oligo(lysine, arginine). The glyoxalase is glyoxalase I or glyoxalase II. The glyoxalase fusion protein has amino acid sequence of sequence number 9 or 11. A pharmaceutical composition for preventing and treating neurodegenerative diseases such as diabetes, Parkinson's disease, and Alzheimer's diseases, and nervous disease such as brain ischemia contains the glyoxalase fusion protein as an active ingredient and pharmaceutically acceptable carrier.

Abstract:
본 발명자들은 PRAS40 융합단백질이 인비트로에서 산화 스트레스로 유도된 신경세포사멸에 대한 세포 보호효과와 인비보에서 뇌허혈 동물모델 보호효과가 있음을 밝혔다. PRAS40 융합단백질은 신경세포 내로 농도 및 시간 의존적으로 효과적으로 투과하였다. 세포 내로 투과된 PRAS40 융합단백질은 과산화수소 독성에 대해 세포생존율을 증가시켰고, 세포내 활성산소종 수준 및 과산화수소로 유도되는 DNA 단편화를 감소시켰다. 동물모델에서는 전뇌 해마 CA1 부분에 일시적 허혈을 유도했을 때 일어나는 신경세포사멸에 대해 PRAS40 융합단백질이 보호효과를 나타내었다. 이러한 결과들은 PRAS40 융합단백질이 세포사멸에 대해 보호효과가 있으며, 뇌허혈 손상으로부터 신경세포를 보호하는 치료효과가 있음을 말해준다.
Abstract:
The present invention relates to a pharmaceutical composition comprising methyl gallate for preventing and treating tumor. Effects of methyl gallate that affect proliferation and metastasis of glutamate-induced rat C6 and human U373 glioma cells are tested. Cell viability and metastasis are significantly reduced when the glioma cells are treated with the methyl gallate compound of the present invention. The result clearly indicates that the methyl gallate has a strong anti-tumor activity by adjusting Akt and ERK1/2 signal transmission path. Therefore, the methyl gallate can be used as an anti-tumor agent for the glioma cells.
