Una composición que comprende: una ADN polimerasa recombinante, comprendiendo dicha polimerasa recombinante una secuencia de aminoácidos que es al menos un 70 % idéntica a la SEQ ID NO:1 y comprende una sustitución de aminoácido en un sitio correspondiente a K512 respecto a la SEQ ID NO:1; y un análogo nucleotídico que está marcado en un grupo fosfato, en el que el resto de base del análogo se selecciona del grupo que consiste en adenina, timina, guanina y citidina.

Methods of making the polymerases and of using the polymerases in sequencing and DNA replication and amplification as well as kinetic models of polymerase activity and computer-implemented methods of using the models are also provided.
