Abstract: GT Systems and methods are disclosed for controlling humidity and/or temperature during chemical analysis of a sample material. Specifically, the present application relates to microfluidics systems and methods, e.g. involving ADE, open port interface (OPI) and/or mass spectrometry (MS), for controlling humidity and/or temperature during chemical analysis of a sample material. The present systems and methods allow a user to modify the temperature of a microplate during dispensing. This allows the user to study reactions that occur at temperatures different than room temperature, e.g. at body temperature. Additionally, modifying and/or controlling the temperature of a microplate during dispensing can allow a user to maintain quality of a sample through maintaining a proper temperature, e.g. a cool temperature to prevent degradation of a sample. As part of the present invention, Applicant determined how to avoid phase changes, e.g. evaporation, that are particularly concerning because of the small amounts of sample involved.

Abstract: An ADE device identifies an identifiable sequence of one or more ejections from at least one sample using a different value or pattern of values for one or more ADE parameters. The identifiable one or more ejections are performed to produce one or more mass peaks that have a different feature value or pattern of feature values for one or more peak features than other mass peaks produced. Ejection times are stored. One or more detected peaks with the different feature values or pattern of feature values are identified as produced by the identifiable one or more ejections. A delay time is calculated from the time of the identifiable ejections and the time of the identified detected peaks and the peaks are aligned with samples using delay time, stored times, and order of the samples.

Abstract: Methods and systems for delivering a liquid sample to an ion source for the generation of ions and subsequent analysis by mass spectrometry are provided herein. In accordance with various aspects of the present teachings, MS-based systems and methods are provided in which the flow of desorption solvent within a sampling probe fluidly coupled to an ion source can be selectively controlled such that one or more analyte species can be desorbed from a sample substrate inserted within the sampling probe within a decreased volume of desorption solvent for subsequently delivery to the ion source. In various aspects, sensitivity can be increased due to higher desorption efficiency (e.g., due to increased desorption time) and/or decreased dilution of the desorbed analytes. The methods and systems described herein can additionally or alternatively provide for the selective control of the flow rate of the desorption solvent within the sampling interface so as to enable additional processing steps to occur within the sampling probe (e.g., multiple samplings, reactions).

Abstract: Methods and systems for delivering liquid sample to an ion source and subsequent analysis by mass spectrometry. In accordance with various aspects of the present teachings, MS-based systems and methods are provided in which desorption solvent is used in sampling interface to desorb analyte species from an SPME device that is coupled to an ion source to ionize analyte species desorbed into the desorption solvent for MS analysis (e.g., without a liquid chromatography (LC) column between the sampling interface and the ion source). In various aspects of the methods and systems described herein, configuring the sampling interface can be optimized so as to reduce the fluid volume dead space about the fluid inlet so as to concentrate the one or more analyte species desorbed at optimized conditions from the SPME substrate in a decreased volume of the desorption solvent when the SPME device is inserted into sampling interface.

Abstract: Provided is the processing of sample fluids containing one or more analytes of interest and to methods and devices for separating and/or purifying components of a sample fluid using electric and hydrodynamic forces. Though the fluid processing systems and methods are generally described herein as applied to microfluidics, it will be appreciated that the fluid processing systems may process any fluid volume suitable for use in embodiments described herein. Y-shaped and multiple-branched shaped 2-D EFD devices have been used to separate and/or purify one or more analytes from a mixture. Systems and methods in accordance with various aspects of the present teachings utilize hydrodynamic pressure (e.g., using a pump) to drive the sample liquid from the sample inlet to the separation stream, and can, in some aspects, provide improved control of the movement of the analytes, improved processing times, and decreased buffer depletion.

Abstract: Methods and apparatus for processing fluids are described. In various aspects, a fluid processing system may include a magnetic assembly that includes a plurality of magnetic structures configured to generate a magnetic field gradient within a fluid container. The magnetic structures may be formed as a plurality of electromagnets configured to be individually actuated by a controller. Each of the electromagnets may generate a magnetic field within the fluid container. The electromagnets may be differentially actuated to create a magnetic field gradient within the fluid container to agitate, mix, or otherwise influence magnetic particles disposed within the fluid container. Activation of the electromagnets of an electromagnetic structure may generate a magnetic field gradient that influences magnetic particles in an x-y direction. In addition, activation of the electromagnets of a plurality of electromagnetic structures may generate magnetic field gradients that influences magnetic particles in an x-y direction and z-direction.
