A sample loader for loading a liquid sample into a plurality of reaction sites within a substrate is provided. The sample loader includes a first blade, and a second blade coupled to the first blade. The sample loader further comprises a flow path between the first blade and second blade configured to dispense a liquid sample to a substrate including a plurality of reaction sites. Further, in various embodiments the liquid sample has an advancing contact angle of 85 +/- 15 degrees with the first and second blade. Furthermore, loading of the liquid sample dispensed from the flow path to the plurality of reaction sites may be based on capillary action.

A method, a computer program and an apparatus for measuring a waveform associated with a chemical event occurring on an ISFET sensor array. The waveform includes at least one region associated with expected measured values and at least one region associated with unpredictable measured values. First frame averaging is applied to at least one region associated with the expected measured values. Second frame averaging is applied to at least one region associated with the unpredictable measured values. The second number of frames used for averaging is less than the first number of frames. By the variable frame averaging the amount of stored data can be reduced.

Compositions and methods for increasing protein recovery and/ or reducing metal ion stripping in IMAC chromatography or from resins used therein are provided. Samples like spent/ conditioned cell culture media or cell extracts containing native or recombinant proteins, were supplemented with metal ions before loading on to IMAC resins or columns to reduce metal ion stripping and/ or to increase protein recovery. The compositions and methods discussed herein work for a variety of metal affinity resins. Use of the compositions and methods disclosed may result in reusability of columns or resins, and improve the yield, purity and cost-effectiveness of using metal-ion based chromatography. The process is scalable to large-scale commercial levels.

The present invention provides methods, compositions, kits, systems and apparatus that are useful for multiplex PCR of one or more nucleic acids present in a sample. In particular, various target-specific primers are provided that allow for the selective amplification of one or more target sequences. In one aspect, the invention relates to target-specific primers useful for the selective amplification of one or more target sequences associated with cancer or inherited disease. In some aspects, amplified target sequences obtained using the disclosed methods, kits, systems and apparatuses can be used in various downstream processes including nucleic acid sequencing and used to detect the presence of genetic variants.

The present invention relates to methods and apparatus for apportionment and manipulation of sample volumes into smaller discrete volumes. The method exploits the interplay of hydrophilic and hydrophobic forces to partition sample volumes. These compartmentalized volumes allow for isolation of samples and partitioning into a localized array that can subsequently be manipulated and analyzed. The partition into extremely small volumes along with the device's inherent portability render our invention versatile for use in many areas, including but not limited to PCR, digital PCR, biological assays for diagnostics and prognostics, cancer diagnosis and prognosis, high throughput screening, single molecule and single cell reactions or assays, the study crystallization and other statistical processes, protein crystallization, drug screening, environmental testing, and the coupling to a wide range of analytical detection techniques for biomedical assays and measurements.

Alkynyl-derivatized cap analogs, alkynyl-modified capped RNA, 1,4-disubstituted triazole-derivatized capped RNA, methods of preparation, methods of isolation, and uses thereof are provided. The "click" modification facilitates detection and isolation of capped RNAs and the 1,4-disubstituted triazole derivatives formed by the "click" reaction are useful for producing RNA transcripts and encoded protein.
