The present invention provides nucleic acid molecules, DNA constructs, plasmids, and methods for post-transcriptional regulation of gene expression using RNA molecules to both repress and activate translation of an open reading frame. Repression of gene expression is achieved through the presence of a regulatory nucleic acid element (the cis-repressive RNA or crRNA) within the 5′ untranslated region (5′ UTR) of an mRNA molecule. The nucleic acid element forms a hairpin (stem/loop) structure through complementary base pairing. The hairpin blocks access to the mRNA transcript by the ribosome, thereby preventing translation. In particular, in embodiments of the invention designed to operate in prokaryotic cells, the stem of the hairpin secondary structure sequesters the ribosome binding site (RBS). In embodiments of the invention designed to operate in eukaryotic cells, the stem of the hairpin is positioned upstream of the start codon, anywhere within the 5′ UTR of an mRNA. A small RNA (trans-activating RNA, or taRNA), expressed in trans, interacts with the crRNA and alters the hairpin structure. This alteration allows the ribosome to gain access to the region of the transcript upstream of the start codon, thereby activating transcription from its previously repressed state.

A telescoping ramp may include first and second ramp sections such that the second ramp section may be received within the first ramp section and translated between an extended position and a retracted position. The telescoping ramp may include a locking mechanism that extends between at least one of pairs of rails of the first and second ramp sections. The locking mechanism may include a pair of locking pins that is selectively engageable with the first and second ramp sections. In one embodiment, the pair of locking pins may include handle portions that may be disposed between the at least one of the pairs of rails. In such embodiment, an axial movement of the handle portions along a common axis of the pair of locking pins may lock the second ramp section relative to the first ramp section.

A closure with a self-resealing elastomeric septum has a shutter slidable between a closed position over the septum and an open position for exposing the septum. The shutter is spring biased towards the closed position, and may be pushed and held away from the closed position with the tip of an oral syringe to gain access for penetration of the elastomeric septum. The closure may have an upper portion mounted to a lower portion fastened for closing a container. A conduit extends through the upper and lower portions to the interior of the container. A break-away connection is adapted to preferentially break or separate when a sufficient bending or twisting force is applied to the upper portion relative to the lower portion. The conduit may be normally closed by a self-resealing elastomeric septum provided in the lower portion and a slidable shutter may be provided on the upper portion.
